Common biological slide making methods

There are three common biological slide methods: freehand slicing, paraffin slicing and collodion slicing:


1. Freehand slicing method: freehand slicing method is simple, time-saving and easy to master. Although it has certain limitations, it is still a basic slicing method for observing morphological structure in biology teaching. Freehand slicing includes the steps of material selection, holding, slicing, taking, selecting and loading.

2. Paraffin sectioning method: the supporting agent used in paraffin sectioning method is paraffin, which has the advantage that the cut pieces are thin and uniform, and can be used for continuous sectioning, but the production procedures are complex, which can be divided into eight steps: fixation, washing, dehydration, dyeing, wax dipping, sectioning, transparency and sealing.

3. Collodion slicing method: collodion is the supporting agent used in collodion slicing method. The advantage of this method is that the embedding process does not go through high temperature, which can avoid tissue shrinkage and embrittlement. It is suitable for slicing of fine materials (such as brain). Also, due to the toughness of collodion, the slicing will not be folded or broken. It is suitable for slicing of large tissue and multi hole tissue (such as brain and eyeball)
