Steps for making biological slides

When we make biological slides from microbial slices of animal specimens, the key point is to prepare the required materials, and then master the sampling method, so that we can make a correct diagnosis. The extraction of materials should be carried out according to the learned theoretical knowledge and operation methods to reduce errors.

The fetching tissue should be kept fresh so that the fixing liquid can penetrate into the tissue evenly. The cutting tissue block should be sharp and can not be turned back during cutting. During cutting, the material should be fetched accurately according to the anatomical part, and the cutting direction should be determined according to the tissue structure of each organ, so as to keep the material clean and maintain the original shape of the tissue.

① wipe the glass slide with gauze;

② Drop a drop of clean water (0.9% normal saline for animal cells) on the clean slide, and the amount of water should not be too much or too little;

③ Take materials from organisms and put them in water droplets;

④ Flatten the torn film and smear the picked material several times to avoid cell overlap and unclear cell mechanism;

⑤ Clamp the cover glass with tweezers to make one side of it contact the water droplets on the slide first (to prevent bubbles), and then slowly cover it on the material to be observed;

⑥ Drop the dye solution on one side of the cover glass;

⑦ Use absorbent paper to absorb the excess dye on the side opposite to the dye drop.


In a word, the biological slide is: take the tissue with moderate size and make biological slices by histological method. Usually, the biological tissue is embedded in paraffin, cut into slices by slicer, then stained with hematoxylin eosin (H-E), and then further examined by microscope to study the development process and make diagnosis.

There are not many preservation requirements for biological slides. Its essence is glass slides, but there is a thin layer of tissue. If the biological slides are kept at room temperature, as long as the biological slides are not broken, this thin layer of tissue does not have to be stored in the refrigerator. If you want to save biological slices by yourself, you can find a small box, put the slices into the box, and store them at room temperature without extrusion or high temperature.
