2. Fixed
Hold one end of the slide in your hand, with the side with the biofilm facing up, and pass the slight fire three times (touch the reverse side of the smear with your fingers, so as not to burn your hands). After the glass slides are cooled, add dye solution.
3. Staining
Put the slide on the slide shelf, add an appropriate amount of ammonium oxalate crystal violet staining solution (or carbolic acid reddening solution) to the film, and dye it for 1~2 minutes.
4. Water washing
Pour out the staining solution, use sterile water in the washing bottle, slowly flow from one end of the slide to the other end, wash away the staining solution, and wash until there is no color of the staining solution in the flowing water.
5. Drying
Use absorbent paper to absorb the water drops on the edge of the smear, and dry it naturally at room temperature or heat it slightly to speed up the drying speed. Be careful not to wipe the bacteria off when using absorbent paper.
6. Microscopic examination
Observe with a microscope and draw the bacterial morphology with a pencil.
